摘要
目的研究微波辐照后大鼠海马组织细胞色素C氧化酶(COX)亚基II、V mRNA表达变化和对酶活性的影响,探讨微波辐照对大鼠海马能量代谢影响的机制。方法采用RT-PCR法检测微波辐照后海马组织COXI、IV mRNA水平,采用还原细胞色素C法和高效液相色谱法(HPIC)分别检测微波辐照后海马组织细胞色素C氧化酶活性和三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、一磷酸腺苷(AMP)含量。结果微波辐照后,大鼠海马组织细胞色素C氧化酶活性和ATP含量明显降低,并与COXII、V mRNA表达变化呈一致性。结论微波辐照可影响大鼠海马COXII、V mRNA表达而降低酶的活性,进而引起能量代谢障碍。
Objective To study mRNA expression of cytochrome c oxidase (COX) subunit I, IV and activity of cytochrome c oxidase in hippocampus of rats after microwave exposure, and to interpret the molecular mechanism of microwave exposure on the energy metabolism deficit in hippocampus of rats. Methods The expression of cytochrome c oxidase subunit Ⅰ and ⅣmRNA after microwave exposure was detected by RT - PCR. The content of adenosine triphosphate (ATP), adenosine diphosphate(ADP), adenosine monophosphate(AMP) and activity of cytochrome c oxidase in hippocampus were detected by high performance liquid chromatography (HPLC) and reduced cytochrome c methods respectively. Results Decreased activity of cytochrome c oxidase and content of ATP in hippocampus were found after exposure, cytochrome c oxidase subunit Ⅰ and Ⅳ mRNA was also downregulated. Conclusion Microwave irradiation can decrease the expression of COXI, Ⅳ mRNA and avtivity of COX, and further to induce energy metabolism deficit in hippocampus of rats.
