鱼鳞胶原蛋白的提取鉴定初探 被引量:18

摘要 胶原蛋白是生物体内含量极高的一种蛋白质,可达30%左右。目前胶原蛋白的主要来源局限于牛和猪的皮和骨,作为下脚料大部分被浪费的鱼鳞也是胶原蛋白的重要来源。采用碱提取、酸提取以及水提取方法提取鱼鳞中的胶原蛋白,并通过SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)方法对提取的胶原蛋白进行鉴定,确定水提法为鱼鳞胶原蛋白的较佳提取方法,并进一步对该提取方法的提取条件进行优化,得出鱼鳞胶原蛋白的最佳提取条件:提取温度70℃,提取时间20min,料液比1:25和1:50之间。 Collagen is the protein that is found in the highest concentration, about 30 %, in the living body. The main sources of industrial collagen are limited to those from bovine and pig skin and bones.Fish scales will have potential as an important collagen source for use in various industries instead of the skins or bones of pig and cattle.Three different extracting methods have been used to study fish scale collagen,SDS-PAGE has also been used to characterize the type of fish scale collagen. Water extracting method was proved to be a better one than the two other methods and was also optimized to figure out the best extracting conditions:extracting temperature is 70 %,extracting time is 20 minutes and the material:fluid ratio is between 1:25 and 1:50.
