摘要
很多植物的新鲜花粉经过Erdtman醋酸酐分解法、洋红等染料和胚胎学制片处理后,形状上会发生很大的变化,这种形态上的变化常常被忽视。本文介绍一种在光学显微镜下,利用油镜油(或纯乙醇)对新鲜花粉进行处理的制片方法。这种方法通过油镜油对花粉覆膜或浸透来改变花粉的透光特性,避免了花粉接触水后吸水膨胀改变形态,使制片不仅可以观察花粉(包括四合花粉)的真实形态、表面纹饰,还能观察到花粉的层次结构。
The pollen preparation, used Erdtman's method or other methods for light microscopic observation, often change the pollen shapes of many plants. The morphological changes are often unaware or omitted by some researchers. The paper gives a simple pollen preparation by using immersion oil (or absolute alcohol) for light microscopic investigation. It makes the pollens, free from the aqueous solution, enclosed with a thin membrane or saturated by the immersion oil. This method can keep their original shapes,and be employed to prepare section and observe the true shape, exterior and layer of the fresh pollen grains (monad and tetrad).
