摘要
目的探讨积雪草甙、苦参碱、川芎嗪等中药有效成分对皮肤瘢痕成纤维细胞生长的影响。方法体外培养瘢痕疙瘩成纤维细胞,分别加入不同浓度的积雪草甙、苦参碱、川芎嗪,通过倒置显微镜观察、MTT法测定细胞活力、乳酸脱氢酶检测等方法分别观测三种中药有效成分对瘢痕成纤维细胞生长的影响。结果与空白对照组相比,实验组中加入积雪草甙0.5mg/ml、苦参碱1.0mg/ml、川芎嗪1.0mg/ml后成纤维细胞数量均明显减少;OD值降低(P<0.05);当积雪草甙浓度高于5mg/ml时乳酸脱氢酶含量超出正常范围。结论有效浓度下,三种中药有效成分均能抑制皮肤瘢痕成纤维细胞生长,其作用强度积雪草甙>苦参碱>川芎嗪。
Objective To analyze the influence of 3 kinds of Chinese herbs(asiaticoside, matrine and tetramethypyrazine) on the growth of fibroblasts of skin scar and synthesis of collagen. Methods The fibroblasts derived from keloid tissue after in vitro culture were divided into a control groups and three trial groups. Asiaticoside,matrine and tetramethypyrazine of various concentrations were added into every culture medium for fibroblasts. The influence of herbal medicine on the proliferation of fibroblasts was observed under inverse microscope, the cellular activity and lactate dehydrogenase activity were also determined. Results Comparing with the control group, the quantity of fibroblasts in the trial groups with the concentration n of 0.5mg/ml asiaticoside , 1.0mg/ml matrine and 1.0mg/ml tetramethypyrazine were obviously decreased and the OD value reduced ( P 〈 0.05), Laetate dehydrogenase contents exceeded normal range when the concentration of asiaticoside reached. Conclusion Under effective concentration, scar fibroblasts proliferation can be inhibited by asiaticoside, matrine and tetramethypyrazine.
