茶碱、咯利普兰对肺纤维化大鼠支气管肺泡灌洗液中炎性细胞的影响

摘要 目的 观察茶碱、咯利普兰对肺纤维化大鼠支气管肺泡灌洗液中炎性细胞数的影响。方法 64只Wistar大鼠随机分为对照组、模型组、茶碱组和咯利普兰组,后三组气管内滴入平阳霉素制备模型,滴入前1d腹腔分别注入生理盐水、氨茶碱溶液[60mg/(kg·d)]和咯利普兰溶液(0.25mg/kg),对照组给予生理盐水;气管内滴药后第7、28天分两批处理动物,收集支气管肺泡灌洗液,进行炎性细胞计数和分类;测定肺匀浆中羟脯胺酸,光镜下观查肺泡炎和肺纤维化程度。结果 BALF中细胞总数和中性粒细胞比例,两治疗组明显低于模型组(P均〈0.05);模型组肺HYP[(2.40±0.15)mg/g肺组织]显著高于荼碱组[(1.42±0.15)mg/s肺组织](P〈0.01)和咯利普兰组[(1.32±0.23)mg/s肺组织](P〈0.01)。结论 茶碱和咯利普兰可减少肺纤维化模型BALF中炎性细胞数,对肺纤维化有阻断作用。 Objective To investigate the effect of theophylline and rolipram on the inflammatory cells in BALF of pulmonary fibrosis model induced by pingyangmycin. Methods Sixty - four wistar rats were randomly divided into four groups:the control group, the model, the theophylline and the rolipram. The latter three were intraperitonea injected with normal saline ( NS), amphylline [ 60mg/( kg·d) ] or rolipram (0.25 mg/kg) each before injected pingyanginycin(5mg/kg) via trachea, and the control injected with NS. At the 7^th and 28^th day after intratrachealed, 8 rats a batch were adminitration to collect samples, which was used to count and classify cell in bronchoalveolar lavage fluid (BALF). The content of hydroxyproline (HYP) was also determined. The left upper lobe was used to histologic study. Results The number of cell and the propotion of neutrophils in BALF in the two therapy groups were lower than those in the model group ( P all 〈 0.05 ). At 28th day the HYP content in the model group[ (2.4±0.15 ) mg/g lung tissue], was higer than the amphulline [ ( 1.15 ±0.15 ) mg/g lung tissue] ( P 〈0.01 ) and the rolipram [ ( 1.32 ±0.23 ) mg/g lung tissue] ( P 〈 0.01 ). Conclusion Amphylline and rolipram can decrease the number of inflammatory cells in BALF of the pulmonary fibrosis model by pingyangInycin,which relieve fibrosis of lung.
