摘要
目的探讨化疗和物理治疗的结合在人回盲肠癌裸鼠肿瘤治疗中的作用。方法人回盲肠癌细胞HCT-8接种到BALB/C雌性裸小鼠腹部皮下,按预期方案进行氟尿嘧啶+高能聚焦超声刀治疗。采用免疫组织化学SP法对瘤组织进行检测。结果成功建立荷瘤小鼠模型。治疗后肿瘤体积无显著变化,局部组织出现焦化,而对照组肿瘤增长明显,无任何外观变化;且治疗后瘤组织中突变型p53蛋白的阳性率有明显下降。结论化疗药物氟尿嘧啶与物理治疗高能聚焦超声刀的有效结合对人回盲肠癌裸鼠肿瘤的治疗有一定效果。
Objective To investigate the effect of 5-Fu combined with focused ultrasound surgery on ileoeecal carcinoma of nude mice. Methods Human ileocecal carcinoma cells (HCT-8) were inoculated into the abdomen subcutis of BALB/C-nu/nu femal nude mice. Fluorouracil(5-Fu) and focused ultrasound surgery were administrated into nude mice according to chemotherapeutic scheme, and the immunohistochemical method was used to examine the effect. Results Tumor bearing model of nude mice was successively prepared. After treatment of 5-Fu and focused ultrasound surgery, the size of tumor was not significantly changed and some eschar could be seen in the tumor, but there was outstanding advance in the control. The expression of mutant type of p53 was decreased obviously in the carcinoma. Conclusion There is a certain efficacy of 5-Fu combined with focused ultrasound surgery in treating ileocecal tumor of mice.
