Expression of granulysin in patients with primary biliary cirrhosis and its clinical significance

摘要 目的:探讨颗粒溶素(granulysin,GNLY)在原发性胆汁性肝硬化和患者(PBC)中的表达及其意义.方法:采用TaqMan探针技术,以18SrRNA为内参照,测定60例PBC患者外周血中GNLYmRNA的含量,同时应用酶联免疫吸附法(ELISA),检测PBC患者血清中GNLY蛋白的水平,并以健康体检组(n=100)和乙型肝炎肝硬化组(n=60)为对照.结果:PBC组GNLYmRNA的平均拷贝数显著高于健康对照组[(2.7±2.5)×108vs(3.0±1.9)×107,P<0.01]和乙型肝炎后肝硬化组[(2.7±2.5)×108vs(4.7±3.6)×105,P<0.001).PBC患者血清中的GNLY蛋白表达显著高于健康对照组(15.48±3.24μg/Lvs4.76±2.32μg/L,P<0.01)和乙型肝炎后肝硬化组(15.48±3.24μg/Lvs2.57±1.84μg/L,P<0.01).结论:GNLY的表达与PBC的发生、发展存在一定的相关性,可用于PBC临床诊断. AIM: To explore the serum expression of granulysin mRNA and protein in patients with primary biliary cirrhosis (PBC). METHODS: Patients with PBC (n = 60), hepatitis B related cirrhosis (n = 60), and healthy controls (n = 100) were included in this study. Based on TaqMan fluorescent probe technique, real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR) was performed to detect the expression of granulysin mRNA, using 18S rRNA as the internal control. The level of granulysin protein was measured by immunohistochemistry. RESULTS: The mean copy number of granulysin mRNA was significantly higher in PBC patients than that in healthy controls [(2.7 ± 2.5) × 10^8 vs (3.0 ± 1.9) × 10^7, p 〈 0.01) or patients with hepatitis B related cirrhosis [(2.7 ± 2.5) × 10^8 vs (4.7 ± 3.6) × 10^5, p 〈 0.001). Meanwhile, the serum level of granulysin protein was also markedly higher in PBC patients than that in healthy controls (15.48 ± 3.24 μg/L vs 4.76 ± 2.32 μg/L) or patients with hepatitis B related cirrhosis (15.48 ± 3.24 μg/L vs 2.57 ± 1.84 μg/L, P 〈 0.01). CONCLUSION: Granulysin expression is associated with the occurrence and progression of PBC, which can help to make diagnosis for PBC.
