新的候选肿瘤抑制基因SLC5 A8的研究进展

摘要 目前研究认为,SLC5A8是一种新的候选肿瘤抑制基因,在人类的结肠癌、胃癌、甲状腺癌、脑胶质细胞瘤及其他一些肿瘤中表达降低或缺失。该基因编码蛋白为一种细胞运载体,属于钠离子/葡萄糖联合载体基因家族(SLC5),间接与钠离子耦联并电离,转运各种简单的羟化物,包括短链脂肪酸、乳酸和烟酸等,可能也转运碘化物。与癌发生相关的基因沉寂、其外显子1的CpG岛的过甲基化和组蛋白去乙酰化相关。作为SLC5A8编码蛋白底物的丁酸是组蛋白脱乙酰基酶的抑制剂,参与介导包括结肠肿瘤在内的多种肿瘤细胞的凋亡。组蛋白脱乙酰基酶抑制剂和甲基转移酶抑制剂为肿瘤的基因治疗提供了方法。
