内皮素1在豚鼠耳蜗外侧壁的分布及意义

摘要 目的 研究内皮素1在豚鼠耳蜗外侧壁的分布,方法选体质量350-400g的健康豚鼠进行实验。给予腹腔注射戊巴比妥麻醉,主动脉插管灌注200ml。生理盐水后,注入40g/L多聚甲醛直到豚鼠颈部僵直,取出颞骨置于40g/L多聚甲醛再固定,10%EDTA脱钙2周,入蜡包埋、切片,以免抗人内皮素1多克隆抗体为一抗,采用免疫组化ABC法及图像分析方法检测内皮素1的表达情况。结果,耳蜗血管纹边缘细胞、螺旋凸细胞,外沟细胞,血管纹内毛细血管,基底细胞,螺旋韧带纤维细胞均见强弱不等内皮素1阳性反应。结论 内皮素1在耳蜗外侧壁广泛分布,可能对耳蜗内环境稳态有重要调节作用。
