顺铂对正常大鼠卵巢颗粒细胞的毒性和金雀黄素的保护作用 被引量:4

摘要 目的研究顺铂(cisplatin,CDDP)对正常大鼠卵巢颗粒细胞的毒性作用及金雀黄素(genistein,GEN)保护作用。方法原代培养的颗粒细胞在含有不同浓度的CDDP及GEN和CDDP的培养基中孵育不同时间,用MTT法测定细胞的抑制率。结果CDDP浓度≥1μg/ml可抑制颗粒细胞的生长,低浓度GEN的对这种抑制无明显影响,高浓度的GEN可减轻顺铂对颗粒细胞的毒性作用。结论CDDP对大鼠颗粒细胞具有明显的毒性作用,GEN对其引起的细胞毒性有保护作用,呈时间-剂量效应。 Objective To explore the toxicity effect of cisplatin on ovarian granulosa cells and the protective effect of genistein. Methods Ovarian granulosa cells of primary culture were incubated with CDDP or GEN and CDDP at different time, Cell inhibition rate was examined by MTT. Results The results showed that CDDP inhibited the growth of ovarian granulosa cells at no less than 1μg/ml. GEN at lower dosage had no effects on the cytotoxicity of CDDP and GEN at high dosage can protect ovarian granulosa cells from the cytotoxicity. Conclusions The results suggest that CDDP had the cytotoxicity on ovarian granulosa cells and genistein may relieve the cytotoxicity induced by CDDP in dose and time manner.
