摘要
目的:探讨CXCR4/SDF-1生物学轴对人乳腺癌细胞MCF-7生长与增殖的影响.方法:采用WesternBlot免疫印迹法检测MCF-7细胞CXCR4蛋白的表达.以CXCR4单克隆抗体(CXCR4mAb)作用MCF-7细胞,通过MTT比色法、软琼脂克隆形成实验和细胞周期检测观察MCF-7细胞的生长情况;通过流式细胞仪检测MCF-7细胞的增殖细胞核抗原(PC-NA)表达.结果:MCF-7细胞表达CXCR4蛋白;CXCR4mAb作用后,MCF-7细胞生长能力明显降低,并且PCNA蛋白含量由对照组的(91.6±5.3)%降为(78.1±3.6)%(P<0.05).结论:CXCR4mAb可明显抑制MCF-7细胞的增殖.
AIM: To study the effect of CXCR4/SDF-1 on growth and proliferation of human breast cancer cell line MCF-7. METHODS: The expression of CXCR4 protein in MCF-7 cells was measured with Western Blot analysis. After the addition of CXCR4 monoclonal antibody ( CXCR4 mAb), the changes of cell growth were observed by MTr, soft-agar colony formation and cell cycle analysis. The expression of PCNA was analyzed by flow cytometry. RESULTS: Western Blot showed that CXCR4 protein was expressed in MCF-7 cells. As compared with control group, the growth rate and the PCNA expression in MCF-7 cells significantly decreased in vitro after the addition of CXCR4 mAb [PCNA: (91.6±5.3)% vs (78.1±3.6)%, (P〈0.05)]. CONCLUSION: CXCR4/SDF-1 may play an important role in suppressing the proliferation of MCF-7 cells.
