摘要
目的探讨arresten对人脐静脉内皮细胞huvec增殖和迁移的影响,了解其抑制血管生成的途径。方法采用MTT比色法,观察不同浓度的arresten对huvec细胞增殖的影响,Transwell小室检测arresten对huvec细胞迁移的影响。结果MTT比色法显示,arresten对huvec细胞增殖具有抑制作用,随着浓度的增加,arresten对huvec细胞增殖抑制作用也增强,但当其浓度增加到800μg·L-1后其增殖抑制率不再提高;arresten浓度为0、400μg·L-1及800μg·L-1时迁移至Transwell小室膜下的huvec细胞数分别为105±8、77±9、53±6(0·05>P>0·01)。结论arresten能够抑制hu-vec细胞的增殖和迁移,这可能是其抑制血管生成的途径之一。
Aim To investigate the effect of arresten on human vascular endothelial cell. Methods MTT was used to measure the proliferation of huvec cells. Transwell was used to measure the migration of huvec cells. Results Proliferation of huvec cells was inhibited by arresten, and with the increase of concentration of arresten, the cell viability of huvec cell lines decreased gradually. When the concentration of arresten was 800 μg·L^-1, the cell proliferation inhibition rate was 64% ± 5%. However, when the concentration of arresten was above 800 μg·L^-1, huvec cells were resistant to arresten. When the concentration of arresten was 0 μg·L^-1, 400 μg·L^-1 and 800 μg·L^-1, cells migrated to the under well were 105 ± 8,77 ± 9 and 53 ± 6 ( P 〈 0. 05 ). Conclusion Arresten can inhibit huvec cells proliferating and migrating, which may be a way that it inhibits angiogenesis.
