海洛因成瘾大白鼠脑细胞凋亡的观察及其机制探讨 被引量:2

摘要 目的:研究海洛因成瘾致脑损害中细胞凋亡现象并探讨其机制。方法:采用递增法人为建立海洛因成瘾大鼠模型,设对照组和模型组,取两组大鼠前额皮质、海马、下丘脑等部位脑组织,运用透视电镜观察和原位末端标记(TUNEL)检测脑组织凋亡细胞。结果:本实验中,在透视电镜下观察到海洛因成瘾大鼠脑内多部位神经元、星形胶质细胞各期凋亡超微病理变化影像,TUNEL检测也显示海洛因成瘾各部位脑组织细胞凋亡数明显高于对照组(P<0·01)。结论:海洛因成瘾大白鼠脑内广泛性出现神经元、星形胶质细胞凋亡。脑组织细胞凋亡参与了海洛因成瘾致脑组织细胞死亡的病理机制。 Objective.. The study on apoptosis in the brain tissues of heroin-addicted rats was made in order to clarify the mechanism of brain lesions which have been made by heroin. Methods: Rats were divided into control group and model group. By application of the method of gradual increase of dosuge, rats were given subcutaneous injection of heroin and the addiction of model was established. The brain tissues of the prefroneal cortex, hippocampus, and hypothalamus from the animals of the two groups were observed by electron microscope and were examined by TUNEL. Result: The ultra-structural pathology's image of apoptosis was found in the neurons and the astrocytes of the brain of heroin-addicted rats. In TUNEL reaction, the number of apoptotic cells of the brain of the model group was larger than the control group ( P 〈0. 01). Conclusion. Apoptosis of the neurons and the astrocytes appeared widely in the brain of heroin-addicted rats. Apoptosis was one of the mechanisms of brain lesions which have been made by heroin.
