依托咪酯对大鼠海马CA1区长时程增强的影响 被引量:2

摘要 目的:观察依托咪酯对大鼠海马CA1区锥体神经元产生的长时程增(LTP)的影响。方法:断头法分离Wistar大鼠海马半脑,用切片机切出400pm厚度的海马脑片,实验分4组。脂肪乳剂LTP组、依托咪酯LTP组、D-2-氨基-5-磷酸戊酸(D-APV)+脂肪乳剂LTP组、D-APV+依托咪酯LTP组的脑片分别以30μl脂肪乳剂、30μl依托咪酯(相当于5μmol/L)、50μmol/L D-APV+30μl脂肪乳剂、50μmol/L D-APV+5μmol/L依托咪酯预孵60min,记录基础兴奋性突触后电流(EPSC)10min,然后给予高频刺激(HFS),继续记录EPSC40min。结果:给予NMDA受体竞争性拮抗剂D-APV后,大鼠海马CA1区HFS不能诱发LTP;脂肪乳剂LTP组给予HFS后,产生LTP,HFS后0~10min、11-40min的EPSC值分别为基础值的154.2%和150.7%;依托咪酯LTP组给予HFS后,EPSC值迅速增加,然后逐渐下降,HFS后0~10min的EPSC值为基础值的127.8%,10min后达基线水平,HFS后11~40min的EPSC值与HFS前差异无统计学意义(P〉O.05)。结论:我们的实验条件所诱发的LTP是NMDA受体依赖型的,5μmol/L依托咪酯不影响大鼠海马CA1区锥体神经元LTP的诱导,但损害LTP的维持。
