Selection of the Mutation Strain with High Productivity by Protoplast Fusion Technique

摘要 以龟裂链霉菌(Sterpomyces rimosus)98#和76#为出发菌株,分别用溶菌酶制得2个亲本的原生质体,其中76#采用15 W紫外灯下30 cm照射25 min,100%灭活,用PEG诱导进行了原生质体的融合.通过比较菌落外观、颜色挑选融合子328株,结合前期代谢情况进一步筛选高单位的融合菌株8株,经反复传代考察融合子的稳定性后得到1株新菌株102#,其发酵单位比亲本提高了13.0%. The protoplasts of strains 98^#and 76^# of Sterpomyces rimosus were obtained by dissolvent enzyme respectively. The protoplasts of strains 76^# were treated with UV light for 15 W, 30 cm and 25 rain, the death rate reached 100 %. The protoplasts fusion was induced by PEG. 328 strains were obtained by selecting the fusion-stains with the character and color of colony, then 8 strains were obtained by further selecting with metabolism, the stability of more progeny was tested and at last one strains 102^# was obtained, the ferment level was improved 13.0 % than parent strains.
