乳腺增生病动物模型的建立及ER、PR表达的观察 被引量:22

摘要 目的了解乳腺增生病模型大鼠的发病机制,观察病理形态及雌激素受体(ER)、孕激素受体(PR)免疫反应阳性细胞表达情况。方法将26只W istar雌性未孕大鼠随机分为对照组(13只)和模型组(13只),采用苯甲酸雌二醇联合黄体酮肌注30 d建立大鼠乳腺增生病模型,对照组仅给予大鼠肌肉注射生理盐水。观察大鼠第2、3对乳房病理组织形态(肉眼、镜下),采用免疫组化法测定大鼠第2、3对乳腺组织中ER、PR表达情况。结果模型组中第2、3对乳头高度显著增高,镜下形态符合乳腺增生症,ER、PR阳性强度、数量随着乳腺增生程度加重而增加。结论大量外源性雌二醇刺激下可使大鼠乳腺组织增生。ER、PR在乳腺中的表达情况与乳腺增生程度密切相关。 Objective To investigate pathogenesis and pathomorphology and estrogen receptor (ER) and progesterone receptor (PR) expression in rat model of cyclomastopathy. Methods Twenty six female nonfertile Wistar rats were divided into control group and model group randomly. The rats of model group were injected with benzestrofol and flavolutan to induce cyclomastopathy, while the rats of control group were injected with normal saline. Pathological and histological examinations were performed on the second and third pairs of breast of the rats. The expression of ER and PR in mammary glandular tissue were analyzed by immuno-histochemistry. Results The second and third pairs of breast of rats were significantly enlarged in model group. The pathology was consistent with cyclomastopathy. The positive intensity and number of ER, PR indicated in immuno-histochemistry were increased while hyperplasia of mammary glands was severe. Conclusion A great quantity of ectogenic estradiol can induce mammoplasia. The expression of ER and PR is correlate with the degree of mammoplasia.
