摘要
目的研究60Co辐照血液淋巴细胞凋亡相关基因Fas/FasL、Bc l-Bax的蛋白表达水平。方法利用淋巴血细胞分离液分离辐照前后血液的淋巴血细胞,应用单色流式细胞术(FCM)检测凋亡相关蛋白的表达。结果经35 Gy剂量的60Coγ射线辐照的全血,淋巴细胞凋亡基因蛋白Fas、FasL表达显著升高,P<0.05,Fas/FasL的比值无显著性差异(P>0.05);而凋亡基因Bc l-2、Bax蛋白表达明显下降,P<0.05,但Bc l-2/Bax的比值下降更加显著,P<0.01。结论60Co辐照可促使离体全血淋巴细胞凋亡,并且可应用凋亡相关蛋白Fas、FasL、Bc l-2/Bax比值作为判断离体全血淋巴细胞经60Co辐照后是否灭活的参考指标。
Objective To investigate the expressions of Fas/FasL,Bcl-2/Bax on the Lymphocytes of blood irradiated by ^60Coγ--ray. Methods We segregated the Lymphocytes of irradiated blood and normal controls by separating medium,the detected the expressions of apoptosis related proteins by flow cytometry (FCM). Results Compared with normal controls , expressions of Fas and FasL on the Lymphocytes of blood irradiated by 35Gy ^60Co γ--ray were improved significantly, P 〈0. 05, but the odds of Fas/FasL was no significance( P 〉0.05). Expressions of Bcl-2 , Bax and the odds of Bcl-2/Bax were decreased, P 〈0.05,but the later was more notable, PSi0.01. Conclusion ^60 Coγ--ray irradiation can make the Lymphocytes of blood in vitro apoptosis , besides , Fas,FasL,Bcl-2/Bax maybe the reference makers judged whether the Lymphocytes of blood irradiated by ^60Coγ-ray in vitro was inactived or not.
