CTGF在PCOS大鼠卵巢中的表达及其作用 被引量:7

摘要 目的应用来曲唑建造多囊卵巢综合征(PCOS)动物模型。观察结缔组织生长因子(CTGF)在PCOS大鼠卵巢中的表达,探讨PCOS发病的可能机制。方法①选择6周龄SD清洁级雌性大鼠40只,随机分为实验组、对照组,每组各20只。应用来曲唑灌胃制备PCOS大鼠模型,在最后一次应用来曲唑24h(并禁食12h)后,缺血法致死,取出卵巢、子宫并称重,②放射免疫法测定血清LH、FSH、E2、P、T水平。③免疫组化法观察卵巢组织中CTGF的表达。结果①动情周期:实验组失去规律变化,提示无排卵。(萤器官重量:卵巢重量实验组显著高于对照组(P〈0,05)而子宫重量显著低于对照组(P〈0,05)。③卵巢组织学检查:实验组可以看到早期发育的较多的小卵泡及闭锁卵泡,囊状扩张卵泡明显增加,颗粒细胞层数减少至2~3层,卵泡内卵母细胞或放射冠消失.黄体数量明显下降并伴有不完全的黄素化,而对照组镜下见多个黄体及不同发育阶段的卵泡、卵泡内颗粒细胞呈多层。多为8~9层。④血清性激素水平:实验组血清T、LH浓度显著高于对照组(P〈0.05),血清E2、FSH、P实验组浓度显著低于对照组(P〈o.05)。⑤在卵泡基底膜、卵巢白膜和血管内皮细胞中,CTGF表达水平实验组显著高于对照组(P〈0,05)。结论①来曲唑诱导的大鼠模型可作为PCOS动物模型。②CTGF可能与PCOS的发生有关。 Objective To set up an animal model for the study of polycystic ovarian syndrome (PCOS) by using the non - steroidal aromatase inhibitor - Letrozole and observe the expression of connective tissue growth factor (CTGF) in the rats with PCOS for investigating the pathogenesy of PCOS. Methods 40 female SD rats of six weeks age were randomly divided into two groups ( 20 rats for each group) : Experiment group and control group. Set up an animal model by using Letrozole. On the day subsequent to last letrozole dose administration, the rats were killed, then uteri and ovaries of the rats were excised. Examined the serum hormone levels and observed histologic changes in ovaries and detected LH ,FSH ,E2 ,P ,T and the expression of CTGF in ovary tissue by using immunohistochemistry. Results ①The estrus cycles of the experiment group showed irregular change, which indicated that the rats had no ovulation. ②The experiment group brought about raising ovarian weight ( P 〈 0. 05) and suppression of uterine weight( P 〈0. 05). ③In experiment group, it is possible to see the early development of little follicles, atretic follicles and the abundance of subcaplular ovarian cyst lined with a thin layer of granulosa cells, h is also can be seen the decreased number of corpora lutea with incomplete luteinization. The control group can be seen more complete corpora lutea and different levels of follicles with thicken layer of granulosa cells. ④In experiment group, although serum estradiol and follicle - stimulating hormone (FSH) and progesterone levels were reduced ( P 〈 0. 05 ), testoterone( T ) levels were elevated as levels of luteinizing hormone ( LH ) were ( P 〈0. 05 ). ⑤In experiment group, the level of the expression of CTGF were obviously elevated in follicular basic membrane, vascular endothelial cell and albuginea of the rats with PCOS( P 〈0. 05 ). Conclusion ① Letrozole - induced model in rats can serve as the model of PCOS. ②CTGF can play an improtant role on the pathogenesy of PCOS.
