摘要
获得高质量的线粒体DNA(m tDNA)是进行m tDNA研究的前提。本研究用碱裂解法和加热法均得到了满足RFLP分析要求的苹果绵蚜m tDNA。但只有碱裂解法提取的m tDNA满足克隆的需要。经分析认为,在实验过程中尽可能的避免核DNA的断裂,是获取高质量m tDNA的关键。
Top-quality mitochondrial DNA are required in studies of mitochondrial genome. MtDNA of woolly apple aphids Eriosoma langerum (Hausmann), which can be used in Restriction fragment length polymorphism (RFLP) analysis, are isolated with both methods of alkaline lysis and calefaction. But if the sequences of mtDNA of woolly apple aphids are to be cloned, only the method of alkaline lysis is suit. In the progress of isolating the mtDNA, avoiding the break of nuclear DNA is the key.
