Expression of P57^(kip2),IGFⅡ,Cyclin E and Caspase-3 and Their Correlation in Gastric Adenocarcinoma

摘要 目的:检测P57kip2、IGFⅡ、Cyclin E和Caspase-3蛋白在胃腺癌组织中的表达、相互关系及意义。方法:采用SP免疫组化法检测73例胃腺癌和20例正常胃黏膜组织中P57kip2、IGFⅡ、Cyclin E和Caspase-3蛋白的表达。结果:73例胃腺癌中P57kip2、IGFⅡ、Cyclin E和Caspase-3蛋白的检出率分别为35.6%(26/73)、47.9%(35/73)、46.6%(34/73)和61.6%(45/73),4种蛋白在正常胃黏膜组织中的阳性率分别为85.0%(17/20)、20%(4/20)、15%(3/20)和90%(18/20)。P57kip2和Caspase-3蛋白在胃腺癌中的阳性表达率显著低于正常胃黏膜(P<0.01,P<0.05),IGFⅡ和Cyclin E在胃腺癌中的阳性表达率明显高于正常胃黏膜(P<0.05,P<0.05);P57kip2蛋白表达与胃腺癌的淋巴结转移有关(P<0.01),Cyclin E蛋白表达与腺癌浸润深度有关(P<0.05),Caspase-3蛋白的表达和腺癌分化程度有关(P<0.05);P57kip2与IGFⅡ、Cyclin E、Caspase-3蛋白表达之间差异有显著性(P<0.01,P<0.05,P<0.01)。结论:P57kip2蛋白低表达可能是胃腺癌发生发展的重要因素,P57kip2可能拮抗Cyclin E蛋白表达,IGFⅡ对P57kip2负向调节,同时P57kip2与细胞凋亡有关。 Objective. To investigate the expression of P57^kip2, IGF Ⅱ , cyclin E and caspase-3 in gastric adenocarcinoma and their relationship. Methods: The expression of P57^kip2, IGF Ⅱ , Cyclin E and Caspase-3 in 73 gastric adenocarcinoma and 20 normal gastric mucosas specimens were detected by SP immunohistochemistry. Results: Positive rates of P57^kip2,IGF Ⅱ ,Cyclin E and Caspase-3 were 35.6%, 47. 9%, 46. 6% and 61. 6% respectively in gastric adenocarcinoma specimens, versus 85.0 %, 20 %, 15 % and 90 % respectively in normal gastric mucosas. The expression of P57^kip2 and Caspase-3 in gastric adenocarcinoma was significantly lower than in normal gastric mucosas(P〈 0.01 ,P〈0.05), while the expression of IGF Ⅱ and Cyclin E in gastric adenocarcinoma was significantly higher than in normal gastric mucosas(P〈0.05, P〈0.05). The correlation was observed between the expression of P57^kip2 and lymph node metastasis(P〈0.01), between Cyclin E expression and depth of invasion(P〈0. 05),and between caspase-3 expression and differentiation(P〈 0.05). Expressions of IGF Ⅱ , Cyclin E and Caspase-3 were significantly related to that of P57^kip2 (P〈0.01,P〈0.05 and P〈0.01 respectively). Conclusion: The down-expression of P57^kip2 protein might be important causes of carcinogenesis and development of gastric adenocarcinoma. And P57^kip2 can suppress the expression of Cyclin E protein. IGF Ⅱ negatively regulates P57^kip2 and P57^kip2 is probably associated with cell apoptosis.
