化学修饰的茯苓多糖抗肿瘤效应的免疫组织化学观察 被引量:19

摘要 目的:揭示化学修饰的茯苓多糖抗肿瘤效应与凋亡相关基因蛋白Fas、Bcl-2和Bax的关系。方法:①在背部皮下接种了Sarcoma 180肿瘤细胞(5×106)的7-8周龄BALB/c小鼠,被随机分为茯苓多糖治疗组和PBS缓冲溶液阴性对照组,每组10只。进行腹腔注射,每天1次,连续注射7 d。于第9天处死小鼠取出肿瘤组织。②用免疫组织化学SP法测定荷瘤小鼠肿瘤组织中Fas、Bcl-2和Bax基因蛋白的表达。结果:实验组Fas阳性率为64.99%,阴性对照组阳性率为4.12%,两组间差异有显著性意义(P<0.01);实验组Bcl-2阳性率为17.97%,阴性对照组阳性率为47.96%,两组间差异有显著性意义(P<0.01);实验组Bax阳性率为48.05%阴性对照组阳性率为2.45%,两组间差异有显著性意义(P<0.01)。结论:化学修饰的茯苓多糖对小鼠Sarcoma 180肿瘤细胞有诱导凋亡的作用。 Objective: To reveal the relationship between the effect of the anti-tumor of the pachyman polysaccharides (PPS) modified by chemical technique and the apoptosis related gene products, such as Bax, Bcl-2 and Fas. Methods: (1) Twenty BALB/c mice at the age of 7-8 weeks, which were inoculated with Sarcoma 180 cells(5× 10^6 cells/ml) at the hypodermis of the back, were assigned into two groups randomly and equally: the group treated by PPS and the control group treated by phosphoric acid buffer solution. All mice were injected intraperitoneally once per day for seven days before were killed at the ninth day and their tumor tissues were harvested. (2) Fas, Bcl-2 and Bax in tumor tissues were assayed by SP. Results: There were significant differences between the two groups in these indices, including Fas, Bcl-2 and Bax expression. Conclusion: The PPS modified by chemical technique can induce the apoptosis of the Sarcoma 180 tumor cells of mice.
