摘要 目的观察金属硫蛋白-1,2(MT-1,2)基因在燃煤型砷中毒患者外周血细胞中的表达。方法选取燃煤型砷中毒患者及非病区正常对照人群各50例,采用实时荧光定量RT—PCR技术测定外周血细胞中MT-1A、MT-1H、MT-2A mRNA的表达。结果与对照组比较,砷中毒组患者外周血细胞MT-1A、MT-1H、MT-2A基因mRNA表达均明显降低,差异有统计学意义(t=33.40、45.64、24.53,P〈0.01)。结论MT在燃煤型砷中毒代谢中有重要的意义,MT mRNA表达降低可能是砷毒性和致癌性的一个易感因素。 Objective To elucidate the molecular mechanism of arsenism caused by coal-burning and to investigate metallothionein (MT-1A, MT-1H, MT-2A) expression in blood cells in coal-burnting arsenism. Methods Blood cell samples were collected from arsenic-exposing patients and health subjects in Guizhou and total RNA was extracted and purified. The expression of three human MT isoforms was determined with real-time quantitative RT- PCR. The β-actin was used as quality control in the real-time quantitative RT-PCR. Results The relative expression of MT-1A, MT-1H and MT-2A was lower in arsenic exposed patients as compared to health subjects controls (t = 33.40,45.64,24.53,P 〈 0.01). Conclusions The MT is of important significance in the metabolism of coal-burning arsenism. The suppression of MT could be a susceptible factor in arsenic toxicity and arsenic carcinogenesis.
