脑胶质瘤中hMLH1、hMSH2基因转录表达的研究 被引量:1

摘要 目的检测脑胶质瘤中hMLH1、hMSH2基因mRNA表达水平,探讨其在胶质瘤发病过程中的作用。方法用半定量RT-PCR法检测42例脑胶质瘤和5例正常脑组织标本中hMLH1、hMSH2基因mRNA的表达情况。结果hMLH1,hMSH2mRNA在正常脑组织中均有表达。在胶质瘤组织中,表达量均低于正常脑组织;hMLH1mRNA表达缺失3例(7.1%),hMSH2mRNA表达缺失7例(16.7%);hMSH2转录表达下降与病理分级相关。结论hMLH1、hMSH2基因在脑胶质瘤中转录表达下降,可能与胶质瘤发病相关。 Objective To investigate into transcriptional expressions ofhMLH1 and hMSH2 in glioma, and analyze the relationship between the etiology of glioma and mismatch repair gene. Methods Transcriptional expressions of the hMLH1, hMSH2 mRNA were examined in 42 glioma tissues and 5 normal cerebral tissues by semi-quantitative RT-PCR. Results hMLH1 and hMSH2 mRNA was all expressed in 5 normal cerebral tissues, while hMLH1 mRNA was not expressed in 3 cases (7.1%) of glioma and hMLH2 was not expressed in 7 (16.7%). Moreover, expression levels of hMLHI and hMSH2 were significantly lower in glioma than in normal tissues, and hMSH2 mRNA express decreasing was correlated with the pathologic grade. Conclusion Low transcriptional expressions of hMLH1 and hMSH2 exist in glioma, which may correlate with the occurrence of glioma.
