土荆芥-水团花对胃溃疡大鼠黏膜保护作用的研究 被引量:13

摘要 目的:通过土荆芥-水团花对一氧化氮(NO)、表皮生长因子(EGF)及表皮生长因子受体(EGFR)的影响,探讨该药对实验性大鼠胃黏膜的保护作用。方法:采用醋酸注射至大鼠浆膜下形成胃溃疡模型,用游标卡尺测量溃疡指数(UI),用硝酸还原酶法测定NO含量,用放免法检测EGF含量,用免疫组化法检测胃黏膜组织EGFR的表达。结果:雷尼替丁组、土荆芥-水团花内容物组,UI,NO及EGF明显低于溃疡生理盐水组(P<0.05);正常大鼠胃黏膜EGFR的表达为弱阳性,定位在胞浆及胞膜,雷尼替丁组、土荆芥-水团花内容物组溃疡边缘组织阳性表达的细胞较正常组和溃疡生理盐水组逐渐增多(P<0.05)。结论:土荆芥-水团花内容物通过促进NO,EGF等胃黏膜的保护因子分泌,通过上调胃黏膜上皮细胞EGFR水平,促进溃疡愈合。 Objective: To study effect and mechanism of Chenopodium-Pilular adina herb (CP) on concrescence of gastric mucosa in experimental gastric ulcerative rats, by changes of the epidermal growth factor (EGF) and expression of the epidermal growth factor receptor (EGFR). Method: To make the ulcer animal model, by inducing acetic acid The index of ulcer (UI) was measured by avemier, the levels of NO and EGF were examined by the method of nitric acid reduetase and radioimmunity, the expression of EGFR in the mueosa around the ulcer was detected by SABC kit immunohistochemieal method and MPIAS mieroimage analysis system. Result: RA group and CP groups compared with NS group, UI, NO, EGF were significant difference (P 〈 0. 05). The expression of EGFR is weakly positive in Normal gastric mueosa cells, which locate to endochylema and cell membrane. EGFR express increased gradually in RA group and CP groups compared with control group and NS group around the ulcer (P 〈0. 05 ). Conclusion: Chenopodium-Pilular adina herb protected gastric mueosa and cured gastric ulcer by Promoting the secretion of NO, EGF, and increasing the expression of EGFR of gastric mueosa endothelial cell.
