摘要 目的观察钙调素依赖蛋白激酶Ⅱ-α(Calcium/calmodulin-dependentProteinKinase-α,CaMKⅡ-α)在阿尔茨海默病(AD)患者海马的表达,了解AD患者海马表达CaMKⅡ-α改变与AD学习记忆丧失的可能关系。方法利用免疫组织化学和体视学方法定量分析10例女性AD患者及年龄、性别等与之匹配的10例老年对照组海马不同区域CaMKⅡ-α神经元数目和染色强度的差异。结果AD海马CA1区CaMKⅡ-α阳性神经元数目显著减少,残存神经元免疫反应性显著增强。结论CaMKⅡ-α的表达在AD海马CA1区选择性受损,这可能与AD学习记忆丧失相关。 Aim To observe the CaMK Ⅱ-α expression changes in the hippocampal subfield of AD patients and the possible relationship with learn memory loss in AD patients. Methods Samples of hippocampus of 10 female AD patients and 10 non-demented female controis matched with age and postmortem delay were obtained from the netherlands brain bank. The number and optical density of CaMK Ⅱ-α expressing neurons in the CA1-CA4 subfields of hippocampus were quantitatively analyzed with immunohistochemical and stereolgical methods. Results CaMK Ⅱ-α containing neurons were selectively lost in the CA1 subfield of AD patients accompanied with the enhanced immunoreactivity of remaining neurons. Conclusions CaMK Ⅱ-α expression was vulnerable in CA1 subregion neurons of AD hippocampus which may be involved in learn and memory loss in AD patients.
