锌对氟致雄性大鼠生殖损伤的拮抗作用 被引量:5

摘要 目的:探讨不同剂量的锌对高氟所致雄性大鼠生殖损伤的拮抗作用。方法:将50只雄性Wistar大鼠随机分为5组,即对照组(去离子水)、氟组(25mg/kg氟化钠)、氟加低锌组(25mg/kg氟化钠+4mg/kg硫酸锌)、氟加中锌组(25mg/kg氟化钠+20mg/kg硫酸锌)、氟加高锌组(25mg/kg氟化钠+50mg/kg硫酸锌),每组10只,灌胃6周。观察氟、锌对精子数量、活动率和畸形率、血清性激素、睾丸生化标志酶及睾丸组织病理学切片的影响。结果:①精子计数、活动率:氟加中锌组大鼠精子计数、精子活动率均高于氟组、氟加低锌组和氟加高锌组,差异均有统计学意义(P<0.05);精子畸形率:氟加中锌组大鼠精子畸形率低于氟组、氟加低锌组和氟加高锌组,差异均有统计学意义(P<0.05)。②血清睾酮含量:氟加中锌组血清睾酮含量高于氟组和氟加高锌组,差异有统计学意义(P<0.05)。③睾丸乳酸脱氢酶活力:氟加中锌组高于氟加高锌组,差异有统计学意义(P<0.05)。睾丸氟含量:氟加中锌组低于氟组,差异有统计学意义(P<0.05)。④睾丸组织病理学检查结果:氟组、氟加高锌组与对照组比较,形态结构疏松;氟加低锌组、氟加中锌组与氟组比较,结构较完整。结论:中等剂量的锌可以拮抗高氟所致的雄性生殖毒性。 Aim :To explore the antagonism of different doses of zinc on male reproductive toxicity induced by excessive fluorine. Methods:A total of 50 male Wistar rats were randomly allocated into five groups:control group, fluorine group, fluorine plus low-dose zinc group,fluorine plus mid-dose zinc group,and fluorine plus high-dose zinc group. The animals were fed with deionized water supplemented with NaF or NaF and ZnSO4 for 6 weeks. The count of sperm and the rates of sperm mobility and aberration were analyzed. The levels of lactate dehydrogenase (LDH) and serum testosterone(T) were examined. Meanwhile the pathological alterations of testes were observed. Results: Compared with fluorine group, fluorine plus low-dose zinc and fluorine plus high-dose zinc group, the counts of sperm and the rate of sperm mobility significantly increased, and the rates of sperm aberration significantly decreased in fluorine plus mid-dose zinc group (P 〈 0. 05). The levels of T in fluorine plus mid;dose zinc group were significantly increased ( P 〈 0. 05 ) compared with fluorine group and fluorine plus high-dose zinc group ( P 〈 0. 05 ). The levels of LDH were significantly higher in fluorine plus mid-dose zinc group than that in fluorine plus high-dose zinc group. The content of fluoride in fluorine plus mid-dose zinc group was significantly lower than that of fluorine group (P 〈 0.05 ). The mierostructure of testis was loosened in fluorine group and fluorine plus high-dose zinc group compared with control group, while the pathological alterations were milder in the fluorine plus low-,mid-dose zinc groups compared with fluorine group. Conclusion: Fluoride can result in reproductive toxicity in male rats, and appropriate zinc can antagonize the toxicity of fluoride.
