摘要
目的:观察不同品系小鼠微卫星位点的多态性。方法:取6-10周龄近交系BALB/C、豫医无毛小鼠、C57、CBA及DBA/2小鼠各5只,颈椎脱臼处死后取新鲜的脑组织0.3-0.5cm^3,提取DNA。随机选择位于小鼠2、3、4和16号染色体上的5对微卫星引物(D2Nds3、D3mit17、D3mit18、D4mit2、D16mit4),应用PCR技术对5种近交系小鼠的DNA多态性进行研究。结果:5对引物在近交系小鼠各基因座上均出现一清晰条带,D16mit4、D3mit17、D3mit18具有多态性,D4mit2、D2Nds3不具有多态性。结论:筛选出的引物D16mit4、D3mit17、D3mit18可用于检测近交系小鼠的品系来源和遗传背景,且所检测的小鼠符合近交要求。
Aim. To study the polymorphisms of microsatellite of inbred strain mice. Methods:Five microsatellites DNA loci on different chromosomes in five kinds of inbred strain mice including BALB/C, Yuyi hairless mice, CBA, C57 and DBA/2 were investigated by PCR analysis with five pairs of primers for D2Nds3, D3mit17, D3mit18, D4mit2, and D16mit4. Results: An allelic gene band was observed with five pairs of primers. Polymorphism was found at D16mit4, D3mit17, D3mit18 loci,but not at D4mit2 or D2Nds3 loci. Conclusion: The screened primers D16mit4, D3mit17, and D3mit18 could be used to detect the origin and genetic background of inbred strain mice.
