摘要
目的:观察低剂量睾酮(T)对心肌梗死后慢性心衰雄性大鼠血清肿瘤坏死因子-α(TNF-α)含量的影响。方法:对120只健康雄性SD大鼠随机行冠状动脉结扎(n=105)或假手术(PS,n=15)。6周后将存活的44只模型鼠随机分为2组:心力衰竭+T治疗组(CHF+T组,n=22,给予T5mg/kg肌内注射)、心力衰竭组(CHF组,n=22,给予安慰剂)。用药12周后测定血清T及TNF-α含量。结果:CHF组大鼠血清T含量明显低于PS组((1.53±0.77)μg/Lvs(4.75±1.61)μg/L,P<0.05),CHF+T组T含量与PS组差异无统计学意义((4.15±0.59)μg/Lvs(4.75±1.61)μg/L,P>0.05)。CHF组较PS组血清TNF-α含量明显升高((8.59±3.22)ng/Lvs(5.29±2.30)ng/L,P<0.05),CHF+T组较CHF组血清TNF-α含量下降((5.97±1.81)ng/Lvs(8.59±3.22)ng/L,P<0.05),与PS组相比((5.29±2.30)ng/L)差异无统计学意义。结论:低剂量T可降低心肌梗死后慢性心衰雄性大鼠血清TNF-α含量。
Aim: To investigate the effects of low dose-testosterone (T) on serum TNF-α level in male rats model of chronic heart failure(CHF). Methods: A total of 120 male SD rats were randomly assigned to perform ligation of coronary artery (n = 105) or pseudo operation (PS, n = 15). Six weeks later, 44 survived model rats were randomly divided into 2 groups: CHF + T group( n = 22) treated with intramuscular injection of T at 5 mg/kg, and CHF group( n = 22) treated with placebo. Serum T and TNF-αlevels were determined after treatmemt for 12 weeks. Results: Serum T level in CHF group was significantly lower than that in PS group( ( 1.53 ± 0.77) μg/L vs (4.75 ± 1.61 ) μg/L, P 〈 0.05 ). Serum T level in CHF + T group had no significant difference from that in PS group( (4.15 ± 0.59) μg/L vs (4.75± 1.61 ) μg/L, P 〉 0.05). Serum TNF-α was significantly higher in CHF group compared with that in PS group ( ( 8.59 ± 3.22 ) ng/L vs (5.29 ±2.30) ng/L, P 〈0.05). Serum TNF-α was markedly lower in CHF +T group compared with that in CHF group ( (5.97 ±1.81 ) ng/L vs (8.59 ±3.22) ng/L, P 〈 0.05). No significant difference of serum TNF-α level was found between CHF +T group and PS group( (5.97 ±1.81) ng/L vs (5.29 ±2.30) ng/L, P〉0.05). Conclusion: Low dose-T can induce a reduction in TNF-α in male rats model of CHF.
