摘要 目的:研究仙鹤草水提液对人食管癌Eca109细胞生长的抑制作用及其可能机制。方法:MTT法测定0g/L、2g/L、4g/L、8g/L、16g/L的仙鹤草水提液分别作用于Eca109细胞24h、48h、72h,观察对Eca109细胞生长的抑制作用;采用免疫组织化学染色技术检测16g/L仙鹤草水提液作用72h对Eca109细胞Bcl-2、P53蛋白表达的影响。结果:MTT结果显示16g/L仙鹤草水提液作用48h和72h后Eca109细胞生长明显受抑;免疫组化结果显示16g/L仙鹤草水提液作用72h后Eca109细胞内Bcl-2蛋白表达下调,P53蛋白表达上调。结论:仙鹤草水提液体外能诱导Eca109细胞凋亡,其机制可能与下调Bcl-2蛋白表达及上调P53蛋白表达有关。 Aim: To study the inhibitory effect of Agrimonia Pilosa Ledeb. (APL) water extract on human esophageal cancer cell llne Eca109 cells. Methods: The effects of APL water extract at 0 g/L, 2 g/L, 4 g/L, 8 g/L, and 16 g/L on the growth and apoptosis of Eca109 cells for 24 h, 48 h, and 72 h were evaluated using MTT. The expression of Bcl-2 and P53 protein was detected using immunohistochemical staining after 16 g/L APL water extract being added to Eca109 cells culture fluid for 72 h. Results: Eca109 cell growth was inhibited dramatically after being treated by 16 g/L APL water extract for 48 h and 72 h. ImmunohistochemicaI staining tests revealed that the expression of Bcl-2 protein was down-regulated, and that of P53 protein was up-regulated after APL treatment. Conclusion : APL water extract could inhibit the proliferation of Eta109 cells in vitro by down-regulating the expression of Bcl-2 and up-regulating the expression of P53 protein.
