前凋亡因子Bim在小鼠胸腺细胞自然凋亡中的作用 被引量:3

摘要 目的研究前凋亡因子Bim在小鼠胸腺细胞自然凋亡中的作用。方法体外培养BALB/c小鼠胸腺细胞不同时间,AnnexinV/PI双染流式细胞术检测细胞凋亡;RT-PCR半定量法分析不同时间点(0,1,2,4,8h)Bim mRNA表达。结果①体外培养的小鼠胸腺细胞随培养时间发生自然凋亡,流式细胞术检测显示随培养时间延长,各时间点细胞凋亡百分比较对照组高(vs Oh,P<0.05);②RT-PCR显示在一定时间内前凋亡因子Bim mRNA随培养时间延长表达增高(vsOh,P<0.05),在Bim 3个异构体中以BimL作用为主。结论Bim参与小鼠胸腺细胞的自然凋亡过程,是重要的细胞凋亡促进因子。 Objective To explore the role of pro-apoptotic protein Bim in the apoptosis of mouse thymocyte culturod in vitro. Methods BALB/c mouse thymocyte was cultured in vitro and detected by AnnexinV-FITC/PI flow cytometry. RT-PCR method was applied to semiquantitative analysis of Bim mRNA expression at different time point(0, 1,2,4,8h). Results ①AnnexinV-FITC/PI flow cytometry analysis showed that apoptosis rate of thymocyte cultured in vitro at different time point ( 1, 2,4, 8h ) raised significantly than control(vs Oh, P 〈 0.05 ) ; ②Bim mRNA expression of mouse thymocyte began to raise after lh and increased significantly than control ( vs 0h, P 〈 0.05 ) and BimL is the predominant isoform in this proces. Conclusion Pro-apoptofic protein Bim plays an important role in mouse thymocyte apoptasis cultured in vitro.
