摘要
目的体外评价维拉帕米对正常牙龈成纤维细胞增殖的影响。方法分离、培养正常牙龈成纤维细胞。用0、0.1、1、10、100 μmol/L 5个浓度的维拉帕米分别作用于第5代正常牙龈成纤维细胞,采用甲基噻唑基四唑法检测细胞的增殖,流式细胞仪评价细胞生长周期。结果 100μmol/L维拉帕米作用66 h 后,检测 A 值为0.046,与无药物作用组(A 值为0.088)相比差异有统计学意义(P<0.01)。100μmoL/L 维拉帕米作用18 h 后,69%的细胞处于 G_0~G_1期,27%处于 S 期,而无药物作用组分别为41%和49%。两者间差异有统计学意义(P<0.001)。结论 100μmol/L 维拉帕米在体外通过阻滞细胞生长周期抑制正常牙龈成纤维细胞的增殖。
Objective To evaluate the effect of verapamil on the proliferation of normal gingival fibroblast(NGF) in vitro. Methods NGF was isolated and cultured. The 5th passage of NGF was incubated with 0,0. 1,1,10,100 μmol/L verapamil respectively. Methyl thiazolyl tetrazolium(MTT) assay and flow cytometry were used to detect cell proliferation and cell cycles. Results Incubated with 100 μmol/L verapamil for 66 h, the A value of normal gingival fibroblast was significantly lower than those without verapamil groups (P 〈 0. 01 ). Incubated with 100 μmol/L verapamil for 18 h, 69% of cells were at the G0-G1 phase, 27% were at the S phase. For control group(without varapamil) 41% of cells were at G0-G1 phase and 49% ceils were at S phase. There was significant difference between the two groups (P 〈 0. 001 ). Conclusions 100 μmol/L verapamil inhibited proliferation of normal gingival fibroblast by a cell-cycle arrest.
