酮色林和坎地沙坦对清醒自由活动自发高血压大鼠压力反射功能的影响

摘要 目的比较酮色林和坎地沙坦对清醒自由活动自发高血压大鼠压力反射功能的影响。方法采用计算机化清醒自由活动大鼠血压监测方法测定血压、血压波动性和心动周期;用Smyth法测定压力反射功能敏感性。酮色林和坎地沙坦均分为1.0、3.0、10.0 mg.kg-13个剂量组。结果酮色林(1.0、3.0、10.0 mg.kg-1)可剂量依赖性地降低清醒自由活动自发高血压大鼠的血压,同时可显著改善其压力反射功能,但改善压力反射功能的作用无剂量依赖性;坎地沙坦(1.0、3.0、10.0 mg.kg-1)可剂量依赖性地降低清醒自由活动自发高血压大鼠的血压,但各剂量组对其压力反射功能均无显著影响。结论酮色林和坎地沙坦均可剂量依赖性降低清醒自由活动自发高血压大鼠的血压;酮色林的改善压力反射功能、稳定血压的作用优于坎地沙坦。 Objective To compare the effect of ketanserin and candesartan on baroreflex in conscious spontaneous hypertensive rats. Methods The blood pressure, variability of blood pressure and heart period of conseious spontaneous hypertensive rats were computerizedly measured. Baroreflex sensitivity was measured by Smyth method. Ketanserin or candesartan was given by 1.0 mg· kg^- 1, 3.0 mg· kg^- 1, 10.0 mg· kg^- 1, respectively. Results Ketanserin(1.0 - 10.0mg· kg^- 1 ) decreased blood pressure in dose-dependent way; and variability of blood pressure was also decreased; Baroreflex sensitivity was improved but not dose-dependent in spontaneous hypertensive rats. Candesartan(1.0 mg· kg^- 1, 3.0 mg· kg^- 1, 10.0 mg· kg^- 1) decreased blood pressure in conscious spontaneous hypertensive rats in dose-dependent way; But it had no effect on baroreflex sensitivity and blood pressure variability. Conclusions Both ketanserin and candesartan can decrease blood pressure in dose-dependent way in conscious spontaneous rats. The effects of decreasing variability of blood pressure and improving BRS of ketanserin are better than that of candesartan.
