更年乐颗粒剂对12月龄大鼠下丘脑-垂体-卵巢轴的调节作用 被引量:3

摘要 目的:通过检测内分泌激素的水平,下丘脑雌激素受体(ER)表达,考察更年乐对下丘脑-垂体-卵巢轴(HPO)的调节作用。方法:采用12-15月龄SD大鼠,同时设青年鼠作对照,灌胃给药30 d后,放免法检测血清E2、P、Te、FSH、LH水平,免疫组化法检测下丘脑GnRH,垂体和下丘脑ER表达。结果:与更年组比,12月龄组可以升高12月龄大鼠血清中E2,P(P<0.05或P<0,01)水平,有降低LH水平的趋势(P>0.05),明显降低下丘脑GnRH的表达(P<0.05),上调下丘脑ER表达。结论:更年乐能改善因体内雌激素水平降低导致的下丘脑、垂体功能的相对亢进状态,上调下丘脑ER表达,增强雌激素效应,调节HPO轴的失调。 AIM : To investigate the effect of Gengnianle Granule on hypothalamic-pituitary-ovarian (HPO) axis in climacteric rats. METHODS : 12-15 months SD female rats were randomized into climacteric group and young control group was selected additionally. After being administrated for thirty days, the level of E2 ,P,Te,FSH ,LH in serum were examined by the radio-immunity method, the expression of GnRH in hypothalame, the expression of ER in hypothalame and pituitary appendage were examined by the immunohistoehemieal method. RESULTS: Compared with climacteric group, Gengnianle Granule could increase the level of E2 ,P in serum (P 〈0. 05 or P 〈0. 01 ), decrease the expression of GnRH in hypothalame (P〈0.05) and had the tendency to decrease the level of LH in serum, could increase the expression of ER in hypothalame. CONCLUSION: Gengnianle Granule could regulate the HPO axis in rat.
