摘要
目的探讨纤维连接蛋白(FN)在肥胖鼠肾组织中的表达。方法24周龄肥胖Zucker大鼠和12周龄肥胖db/db小鼠用于实验,以瘦Zucker大鼠和瘦db/m小鼠分别作为对照组。采用RT-PCR和免疫组织化学方法分别检测FN在肾组织中的表达。结果肥胖db/db小鼠的体重和血糖明显高于对照组db/m小鼠(P<0.01)。肥胖Zucker大鼠的体重明显高于对照组瘦Zucker大鼠(P<0.01),但肥胖Zucker大鼠的血糖未明显高于对照组瘦Zucker大鼠。RT-PCR结果表明肥胖db/db小鼠和肥胖Zucker大鼠肾皮质FN mRNA的表达明显高于各自的对照组(P<0.01)。病理检查表明肥胖db/db小鼠和肥胖Zucker大鼠肾小球明显大于各自的对照组,肾小球内FN的表达也明显高于各自的对照组。结论单纯肥胖可引起肾脏纤维化。
Objective To investigate the expression of fibronectin (FN) in obese rats' kidney. Methods 24-week-old male obese Zucker rats and 12-week-old obese db/db mouse were used in this study, with lean Zucker rats and lean db/m mouse as controls respectively. RT-PCR and immunohistochemistry methods were used to detect FN mRNA and protein expression in the renal cortical tissue respective- ly. Results Compared to the lean controls, obese db/db mouse showed significantly increased body weight and serum glucose level(P 〈 0. 01 ). Similarly, compared to the lean controls, obese Zucker rats showed significantly increased body weight( P 〈0.01 ), but not serum glucose level. RT-PCR indicated the expressions of FN mRNA were increased in both obese Zucker rat and db/db mouse cortical tissue than those in their matched controls(P 〈0.01 ). Pathological data showed glomeruli size in obese Zucker rat and db/db mouse was lager than those in their matched controls. Furthermore, the expression of FN protein also increased in glomeuri of obese Zucker rat and db/db mouse than those in their matched controls. Conclusions Simplex obesity can induce renal fibrosis.
