摘要
目的观察米非司酮对子宫腺肌病腺肌组织细胞的作用。方法采用MTT法观察米非司酮对11例子宫腺肌病患者腺肌组织原代培养细胞体外生长状态的影响,实验分为1.25mg/L、2.5mg/L、5mg/L 3个药物浓度组,培养52h,测定OD值,并与空白对照组对比。结果5mg/L、2.5mg/L、1.25mg/药物浓度组及空白组的OD值分别为0.219±0.044、0.185±0.038、0.152±0.033、0.262±0.0678,5mg/L与1.25mg/L组有显著差异(P<0.01),5mg/L、2.5mg/药物浓度组与空白组有显著差异(P<0.01)。细胞生长抑制率2.5mg/L组(IR=29.4%)是1.25mg/L组(IR=16.4%)的1.79倍,5mg/L组(IR=42.0)是2.5mg/L组的1.43倍。结论米非司酮能够抑制子宫腺肌病腺肌组织细胞的生长,其抑制能力与药物浓度呈正相关。
Objective: To observe the effects of mifepristone on adenomyosis cells. Methods: Using MTT assay, the growth state of primary cell culture from tissues of 11 cases of adenomyosis was detected, The cells of each case were divided into four groups, three dosage groups were 1.25mg/L, 2.5mg/L, 5mg/L mifepristone respectively and one control group and surveyed their OD value through 52 hours culture. Results: By contrasting the results, the OD values of 1.25mg/L, 2.5mg/L, 5mg/L and control group were 0. 219 ± 0. 044, 0. 185 ± 0. 038, 0. 152 ± 0. 033 and 0. 262 ± 0. 0678 respoefivdy. The OD value of 5mg/L group had significant difference from that of 1.25mg/L group ( P 〈0. 01 ) and 5mg/L, 2. 5mg/L group had signaificant difference from the control group ( P 〈0.01). The inhibition rate of cell growth showed that 2. 5mg/L group (IR =29.4% ) was 1.79 times 1.25mg/L group (IR = 16. 4% ), and 5mg/L group ( IR =42. 0% ) Was 1.43 times 2. 5mg/L group. Conclusions : Mifepristone could suppress the growth of adenomyosis cells in vitro. There was positive correlations between the inhibition growth ability and the mifepfistone concentration.
