中药天年饮对衰老大鼠睾丸生精细胞凋亡及相关因素的影响 被引量:3

Effects of Traditional Chinese Medicine Tiannianyin on Apoptosis and Related Factors of Spermatogenic Cells in Testis of Aging Rats

摘要 目的研究中药天年饮对衰老大鼠睾丸生精细胞凋亡及相关因素的影响。方法40只SD大鼠随机分为4组,正常组、衰老模型组、天年饮用药组及阴性对照组,每组均为10只。D-半乳糖连续腹腔注射制作亚急性衰老的大鼠模型。采用ELISA方法检测各组大鼠血清睾酮含量、SP免疫组化法检测睾丸生精细胞p53蛋白的表达情况、TUNEL法检测各组大鼠睾丸生精细胞的凋亡情况。结果衰老大鼠血清睾酮含量明显降低(模型组与正常组比较:P<0.05),衰老大鼠睾丸p53阳性细胞率及凋亡指数明显高于正常大鼠(P<0.01)。天年饮用药组大鼠血清睾酮含量较衰老大鼠明显升高(P<0.05),天年饮用药组大鼠睾丸p53阳性细胞率及凋亡指数较衰老大鼠明显降低(P<0.05,P<0.01)。结论中药天年饮可改善衰老大鼠的生精功能,具有一定延缓衰老的作用。 Objective To observe the effects of traditional Chinese medicine Tiannianyin on apaptosis and related factors of spermatogenic cells in testis of aging rats. Methods 40 SD rats were applied in this study and divided into 4 groups at random : normal group, aging model group, Tiannianyin group, negative control group, and each group had 10 rats. The sub -acutely aging model rats were made by injecting D - gal into abdominal cavity continuously. ELISA was used to detect the testosterone content in serum, SP immunohistochemistry to observe the p53 expression and TUNEL to detect apaptosis of spermatogenic cells in testis. Results The testosterone content in serum of aging rats decreased significantly compared with normal rats ( P 〈 0.05) ; the p53 positive cells and apoptotic index of aging rats were much higher than those in normal rats ( P 〈0.01 ). Tiannianyin could inhibit the decrease of testosterone content in serum significantly (Tiannianyin group compared with aging model group: P 〈 0.05), and decrease the p53 expression and apaptotic index significantly( P 〈 0.05, P 〈 0.01 ). Conclusion Traditional Chinese medicine Tiannianyin can improve the spermatogenic function of aging rats, and has fairly anti - aging effects.
