摘要
目的为进一步研究我们克隆到的大鼠新基因Rtap2a,本研究采用多种生物信息学方法对该序列进行特征分析和功能预测,以获得该基因及其编码蛋白更多的功能提示。方法应用B1ast、ORFFinder、ScanProsite、TMpredServer和UniGene等软件或数据库,进行序列的相似性比较,开放读码框预测、染色体定位、电子表达谱和编码蛋白质的功能等分析。结果我们得到的Rtap2a序列是该基因的全长cDNA序列,在多物种间高度保守,该基因编码的蛋白可能为一N端在胞内的单次跨膜蛋白;其小鼠同源基因的电子表达谱提示Rtap2a特异地高表达于睾丸组织,在成年小鼠的睾丸中表达量最高。结论大鼠新基因Rtap2a是一睾丸限制性表达的新基因,其编码蛋白单次跨膜,进化上高度保守,可能在哺乳动物的睾丸发育和精子发生过程中具有重要功能,也有可能参与肿瘤的发生、发展等过程。因此,新基因Rtap2a有进一步研究的价值。
Objective: A rat testis eDNA expression library was screened to look for the xenogeneie homologousmolecules.Then a novel rat cDNA Rtap2a was found.d.uring the.screen.and cloned. Bioinformatic analysis was performed in order tostudy the possible function of this cDNA sequence. Methods: Various bioinformatic methods such as Blastn、ORF Finder,ScanProsite、TMpred Server and UniGene were used to analyze and predict the feature and possible functions of the novel cDNA scqoence.Results:Rtap2a was the full-length of a novel rat gene which may have conservative and basic functions in life.There was a single strong transmembrane helix withN-terminus inside predicted by TMpred Server.The homologous mouse Unigene analysis showed Rtap2a was specifically expressed in testis tissue ,and highly expressed in adult mouse testis.Conclusion:The novel gene Rtap2a is evolutionally conserved,restrictedly expresses in testis,and contains a single transmembrane region.It may function in spematogenesis and testis development and play a role during the tumor genesis and development.Theerefore,the functions of the novel gene are valuable for further investigations.
