番茄子叶、下胚轴植株再生体系的建立 被引量:15

THE ESTABLISHMENT OF PLANT REGENERATION FROM COTYLEDON AND HYPOCOTYL EXPLANTS OF TOMATO(Lycopersicon esculentum Mill.)

摘要 本研究以番茄(Lycopersicon esculentumM ill.)栽培品种“中杂9号”、“毛粉802”和“合作908”为试材,通过对番茄子叶、下胚轴的离体培养,研究不同基因型以及不同激素配比对植株再生的影响。结果表明:以下胚轴为外植体时3种材料均可诱导再生植株,其中中杂9号和毛粉802最适宜诱导愈伤组织和芽分化的培养基为MS+6-BA2.0mg/L+IAA0.5mg/L,合作908最适宜诱导愈伤组织和芽分化的培养基为MS+6-BA2.0mg/L+IAA0.4 mg/L。以子叶为外植体时毛粉802和合作908均可诱导出再生植株,其中毛粉802诱导愈伤组织及芽分化的最适宜培养基为MS+6-BA1.0mg/L+IAA0.2mg/L和MS+6-BA1.0mg/L+IAA0.5mg/L,合作908诱导愈伤组织及芽分化的培养基为MS+6-BA0.5mg/L+IBA0.5mg/L。两种外植体适宜的生根培养基均为MS+IAA0.5 mg/L。 Tomato (Lycopersicon esculentum Mill. )cultivars ,Zhong - za No 9 , Mao - fen 802 and He - zuo 908 were used as the materials, and cotyledon and hypocotyl were cultured to study the effect of genetype and different hormone constitution on plant regeneration. The results showed that three eultivars could induce plant regeneration with hypoeotyl as explant, Th best callus initiation and shooting medium was MS + 6 - BA2.0mg/L + IAA0.5mg/L for Zhong - za No 9 and Mao - fen 802, MS + 6 - BA2.0mg/L + IAA0. 4mg/L for He - zuo 908. Mao - fen 802 and He - zuo 908 could induce plant regeneration with cotyledon as explant ,the beat shooting mediums were MS + 6 - BA1.0mg/L + IAA0.2mg/L and MS + 6 - BA1.0mg/L + IAA0.5mg/L for Mao - fen 802. The best shoot- ing medium was MS + 6 - BA0.5mg/L + IBA0.5mg/L for He - zuo 908. MS + IAA0.5mg/L was the best rooting medium for both the explants.
