摘要 目的:研究糖基化蛋白诱导培养下,糖肾汤对大鼠肾小球系膜细胞表达基质金属蛋白酶组织抑制物1、2(TIMP1、TIMP2)mRNA的影响。方法:采用血清药理学及体外大鼠肾小球系膜细胞培养方法,用糖基化牛血清白蛋白(AGE-BSA)刺激肾小球系膜细胞,采用RT-PCR法检测系膜细胞TIMP1、TIMP2 mRNA表达。结果:糖基化蛋白可上调TIMP1、TIMP2 mRNA表达,糖肾汤能降低TIMP1、2mRNA表达。结论:糖肾汤对金属蛋白酶系统紊乱有一定的调节作用。 Objective: To discuss the effects of Tangshen Decoction on mRNA expressions of TIMP1 and TIMP2 in rat mesangial cell under the induction and cultivation of glycosylated protein. Methods: Serum pharmacology and vitro cultural method of rat mesangial cell were adopted, and the glycosylated protein (AGE-BSA) was used to stimulate mesangial cell. Then the mRNA expressions of TIMP1 and TIMP2 in rat mesangial cells were detected by reverse transcription - polymerase chain reaction (RT-PCR) method. Results: Glycosylated protein can up-regulation the mRNA expressions of TIMP1 and TIMP2, and Tangshen Decoction can decline the mRNA expressions of TIMP1 and TIMP2. Conclusion: Tangshen Decoction can adjust the disorder of the tissue inhibitor of metalloproteinase system.
