花粉管通道法对茶树进行dsTCS基因转化的初步研究 被引量:9

摘要 以祁门槠叶群体种茶树植株为受体材料,采用花粉管通道法将茶树咖啡碱合成酶dsRNA(dsTCS)注射到茶花子房中。试验共处理茶花500朵,收获T0种子43粒,加温催芽后出苗38棵,随机选取15棵处理后的茶株和5棵对照茶株,剪取相同部位幼叶,用HPLC测定其咖啡碱含量。检测结果为,处理后的15棵茶株,其平均咖啡碱含量均低于对照茶株,初步表明经花粉管通道法导入的dsTCS部分抑制了茶树体内咖啡碱的合成。 With tea plants of keemenzhong as reporters, tea caffeine synthase dsRNA (dsTCS) was transferred into 500 flowers of receptors through pollen-tube pathway. Forty-three seeds from transferred receptors and 38 plants were obtained. With seed-setting rate of 8.6%, 15 of treated tea plants which were transferred into dsTCS and 5 untreated tea plants were selected, with 2 seeding leaves cut from the same segment of each plant for the measurement of caffeine. The result is the proportions of caffeine in all of the 15 single tea plants are lower than that in the untreated ones, with the most obvious one lower by 44. 01% ,which proves that transformed dsTCS refrain the synthesis of caffeine in tea plants.
