摘要 目的:建立反相高效液相色谱法测定脱氧核苷酸钠及其制剂含量.方法;采用Kromasil C18色谱柱,流动相为0.02mmol·L^-1磷酸盐缓冲液(pH5.2),流速1.0ml·min^-1,柱温36℃,检测波长260nm。结果:脱氧核苷酸钠中dAMP、dGMP、dCMP、dTMP均基线分离,在250.0~15.6ug·ml^-1范围内。相关系数均为0.99914种脱氧核苷酸钠回收率在100.3%~101.1%之间。结论:该方法快速、简便、准确,制荆中辅料无干扰,可用于该药物原料及其制荆的含量测定。 Objective. To establish a HPLC method for determining the Sodium Deoxyribonucleotide and its preparation. Methods. The HPLC was performed on a column of Kromasil C18 (0. 5 um, 4. 6 × 200 mm) at 36℃ ,using phosphate buffer solution(pHS. 2), and the detection wavelength of 260 nm. Resuts: The contents of Sodium Deoxyribonucleotide can be determined and baseline separated. A good linearity were obtained over the range of 250.0-15.6 ug · ml^-1,and the correlation coefficient of four deoxyribonucleic sodium were 0. 999. The recoveries of Sodium Deoxyribonucleotide added were 100. 3%-101. 1%. Conclusion. The method was proved to be simple, rapid and could be used for determination of deoxyribonucleic sodium and its preparation.
