摘要
目的研究DNA氧化损伤修复相关基因hOGG1和MGMT在正常人支气管上皮细胞(Human bronchial epithe-lial,16HBE)、反式7,8-二羟-9,10-环氧苯并芘(BPDE)转化细胞和裸鼠成瘤细胞中的表达。方法采用原位杂交技术检测hOGG1和MGMT在正常16HBE、反式BPDE转化细胞和裸鼠成瘤细胞中mRNA水平的表达。结果hOGG1、MGMT mRNA在反式BPDE转化细胞中阳性表达明显增强,而在裸鼠成瘤细胞中阳性表达相应减弱。结论DNA损伤修复相关基因hOGG1、MGMT mRNA在转化细胞模型中的表达水平明显增强。
Objective To detect the expression of repair genes hOGGI and MGMT related to DNA oxidative damage in 16HBE, anti-BPDE transformed cells and carcinoma cells induced by anti-BPDE transformed cells in nude mice. Methods In situ hybridization(ISH) assay was used to detect the expression of hOGGI mRNA and MGMT mRNA in the 3 above mentioned cell lines respectively. Results The expression of hOGG1 、 MGMT mRNA were increased in trans-BPDE transformed cells, while decreased in carcinoma cell induced by anti-BPDE transformed cells in nude mice. Conclusion The expression of repair genes hOGG1, MGMT related to DNA oxidative damage could be higher in lung cancer anti-BPDE transformed cell models.
