An overview of matrix metalloproteinases in the study of acute pancreatitis

摘要 基质金属蛋白酶(MMPs)是细胞外基质代谢过程中起主要作用的一组蛋白酶类。在急性胰腺炎中,MMPs表达增加,其表达程度与病情密切相关。应用MMPs抑制剂处理后,胰腺组织的局部炎症、坏死程度和继发性损伤明显减轻。MMPs可通过炎症组织的中性粒细胞表达,多种细胞因子能刺激中性粒细胞增殖,进一步促进MMPs释放,破坏基底膜,造成局部和远隔器官损害。此外,MMPs还参与了急性胰腺炎的组织修复过程。 Matrix metalloproteinases (MMPs) play a major role in the metabolic process of extracellular matrix. Inappropriate activation of MMPs mediate significant steps in the pathogenesis of acute pancreatitis. MMPs are detected in a high percentage in acute pancreatitis, in contrast to the absence in most normal condition. In acute pancreatitis, considerable correlations have been detected between MMPs and severity of the pathogenetic condition. In the cases treated with MMPs inhibitors, the inflammation, necrosis and secondary lesions are significantly relieved. As neutrophils of the inflammatory architectures express MMPs, and many cytokines stimulate the proliferation of neutrophils, thus many of these cytokines can facilitate the release of MMPs, dissolve the basal membrane of endothelial cell, and cause damages. In addition, MMPs participate in the process of tissue reparation in acute pancreatitis.
