柱前衍生RP-HPLC法测定桑叶中氨基酸含量 被引量:6

摘要 以邻苯二甲醛(OPA)与9-芴基羰酰氯(FMOC-Cl)作衍生剂,正缬氨酸为内标利用二极管阵列(DAD)可变波长检测程序,采用柱前衍生反相高效液相色谱法(RP-HPLC)测定5个桑叶品种的氨基酸含量。结果表明,各种氨基酸的保留时间(Rt)、相对峰面积(Ax/AI)的RSD均较满意,回收率为93.21%~114.24%;5种桑叶品种中含有大量的氨基酸,含量较高的是异亮氨酸(Ile)、谷氨酸(Glu)、天冬氨酸(Asp)、亮氨酸(Leu)和丙氨酸(Ala),占总含量的60%以上。5种桑叶品种的氨基酸含量从大到小为:南2、粤诱36、粤诱18、粤诱32、粤诱87。该方法适合桑叶种氨基酸含量的测定。 To study the contents of amino acids of the samples of mulberry leaves from five varieties ,the amino acids of mulberry leaves were derived with o-phthaldialdehyde (OPA) and 9-fluorenylmefloyl chloroformate (FMOC-Cl) in precolumn,and DAD variable-wavelength as measure function,and separated by RP-HPLC.Results showed that the RSD for retention time (Pt) and Ax/At of amino acids were satisfied respectively,and the reclaim ratio was 93.21%- 114.24 %. The main amino acids of five mulberry leaves varieties were Ile, Glu ,Asp, Leu and Ala,which occupied above 60 % of total amino acids.Among five mulberry leaves varieties,the content of total amino acids from high to low was Nan2,Yueyou36,Yueyoul8,Yueyou32 and Yueyou87.This method was suitable for the determination of amino acids in mulberry leaves.
