摘要
目的探讨血塞通注射液对大鼠心肌缺血再灌注损伤的保护作用。方法将24只Wistar大鼠随机分为三组:心肌缺血再灌注组(MIR,n=8):术前给予生理盐水干预;血塞通治疗组(X,n=8):术前给予血塞通注射液600mg/(kg.d)腹腔注射给药3天;假手术组(S,n=8):术前处理同MIR组。MIR和X组大鼠在结扎左前降支30分钟后行再灌注240分钟。S组所有操作同MIR组,大鼠仅穿线不结扎左前降支。分别于缺血再灌注240分钟采血,S组取相同时点采血。检测血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)和肌钙蛋白I(cTnI)含量变化。结果与X组和S组比较,MIR组血清SOD活性显著降低(P<0.001),MDA、cTnI含量显著升高(P<0.001);与S组比较,X组血清SOD、MDA浓度差异无显著性(P>0.05),而血清cTnI显著升高(P<0.01)。结论血塞通可能通过减少MDA的产生,增加SOD活性,减轻心肌缺血再灌注损伤。
Objective To investigate the protective effects of Xuesaitong injection on myocardial ischemia-reperfusion injury in rats. Methods 24 wistar rats were randomly divided into three groups:myocardial ischemia-reperfusion group (MIR) that pretreatment with saline for 3 days before operation; Xuesaitong treated group (X), pretreatment with Xuesaitong injection 600mg/kg per day for 3days; Sham group (S) that pretreatment same as MIR group. Xuesaitong and saline were injected into the abdominal cavity. Rats of MIR and X were subjectived to 30 min of left anterior descending coronary occlusion then reperfusion with hemodynamic: reperfusion 240 min in MIR and X rats. In S group rats the left anterior descending coronary was not ligated but the myocadic was penetrate through with thread and other operations like MIR group. The activities of superoxide dismutase (SOD), the content of malondialdehyde (MDA) and cardiac Troponin Ⅰ (cTnⅠ) of serum in MIR, X and S group were observed. Results The levels of serum SOD in MIR group was significantly lower than those in X and S group (P〈0. 001). The levels of serum MDA,cTnⅠ in MIR group were significantly higher than those in X and S group (P〈0. 001). The levels of serum cTnⅠ in X group was remarkably higher than S group (P〈0. 01). Conclusion Xuesaitong possess protective effects on myocardial ischemia-reperfusion injury in rats.
