摘要
目的研究不稳定型心绞痛(UAP)患者血清中IL-6、TNF-α和T淋巴细胞IFN-γ(TH1)、IL-4(TH2)的表达在发病过程中的作用。方法25例发病期和缓解期UAP患者血液标本以及20例正常健康对照标本血清IL-6和TNF-α用放免法检测,外周血T淋巴细胞IFN-γ和IL-4的表达用流式细胞仪四色分析技术检测。结果经配对资料t检验,发作时患者血清中IL-6(762.8±318.4)pg/ml、TNF-α(3.1±1.6)ng/ml,外周血T淋巴细胞IFN-γ(14.8±2.4)%,均显著高于正常对照[分别为(402.6±196.4)pg/ml、(2.0±0.5)ng/ml、(8.8±2.4)%]和缓解时[分别为(462.4±216.8)pg/ml、(2.1±0.4)ng/ml、(9.4±3.0)%],P<0.01。患者发病时淋巴细胞表达IL-4(2.6±1.4)%与正常对照组(2.2±1.4)%和缓解时(2.4±1.5)%比较均无差异(P>0.05)。结论UAP患者发病过程中IL-6和TNF-α显著升高,同时表现TH1细胞功能亢进。
Objective To research the effects of eytokines on pathogenesis of unstable angina peetoris (UAP) by detecting the levels of interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha) in serum and the expressions of interferon gamma (IFN-gamma) and interleukin-4 (IL-4) in T Cells of the patients with UAP. Methods The blood samples from 25 patients with UAP and 20 cases of heahhy controls were detected. The concentrations of IL-6 and TNF-alpha in serum were determined by RIA. The cytokine-producing T cells were quantified by four-color flow eytometry. Result The levels of IL-6 (762.82±318.41 pg/ml) and TNF-alpha ( 3.11 ± 1. 63 ug/ml) in serum and IFN-gamma-producing T cells ( 14.78 ± 2.36 % ) were significantly higher in the patients with onset of UAP than in those during recovered period (462.41 ± 216.82 pg/ml and 2.12 ±0.42 ng/ml, 9.40 ± 3.02% ) and the control group (402.60 ± 196.35 pg/ml and 2.02 ± 0.54 ng/ml, 8.78 ± 2.37% ), but no significant difference was found for IL-4-producing T cells ( P 〉 0.05). Conclusion The cytokines may play important roles in onset of UAP, and imbalance of Th1/Th2 (up-regulation of Thl ) exists in the patients with UAP.
