摘要
【目的】建立库仑阵列电化学高效液相色谱检测法测定大鼠脑组织匀浆中的单胺类神经递质的含量。【方法】色谱柱:ESAHR-80,4.6×8cm;流动相为:一水柠檬酸、柠檬酸三钠、1-庚烷磺酸钠、乙二胺四乙酸二钠、甲醇的混合液。采用美国ESA公司的高效液相色谱-库仑4通道电化学检测器检测去甲肾上腺素(NE)、3,4-二羟基苯乙酸(DOPAC)、多巴胺(DA)、高香草酸(HOMV)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)。【结果】最低检测限为0.15~1.67μg/L。平均回收率为78.68%~105.53%。【结论】本方法操作简便,准确,快速,灵敏度高。可用于天鼠神经精神疾病模型有关神经递质的检测和研究。
[Objective]To establish a measurement method on determining the level of monoamines neurotransmitters and their metabolite8 in rat cerebral homogenate by high performance liquid chromatographic analysis with Coularray Detector-4. [Methods]Column: ESA HR-80,4.6 × 8 cm, Mobile phase: Citric acid trisodium salt, Citric acid monohydrate, 1-Octanesulfonic acid sodium salt and EDTA-Na2, methanol mixture. High performance liquid chromatographic analysis with Coularray Detector-4 was made by ESA Company from U. S. A. The levels of NE, DOPAC, DA, 5-HT and 5HIAA from rats cerebral homogenate were determined. [Resulits]The minimum measurement limit was 0. 15~1.67 μg/L. The average recoveries were 78. 68% ~ 105. 53%. [Conclusion]This method is easy to operate, rapid, accurate and highly sensitive, and could be applied in quantity determination and study of neurotransrnitters in neuro-psychiatrie disease's rat model.
