脑啡肽对培养新生大鼠海马胶质细胞白介素1α基因表达的影响 被引量:1

摘要 用逆转录-聚合酶链反应技术研究了脑啡肽对培养的新生大鼠海马胶质细胞IL-1α基因表达的影响.结果发现,脂多糖(10mg·L-1)能诱导海马胶质细胞IL-1α基因表达,当预先用甲硫脑啡肽(0.1nmol·L-1)或亮脑啡肽(0.1nmol·L-1)处理海马胶质细胞,则脂多糖诱导的海马胶质细胞IL-1α基因表达受到抑制,且脑啡肽的作用可被阿片受体阻断剂纳洛酮所阻断. The influence of enkephalin on IL 1α gene expression induced by lipopolysaccharide (LPS) in cultured hippocampal gliacytes of neonatal rat was investigated by the reverse transcription polymerase chain reaction technique. The results showed that LPS (10 mg·L -1 ) could induce IL 1α gene expression in hippocampal gliacytes. When the hippocampal gliacytes were pretreated with Met enkephalin (0.1 nmol·L -1 ) or Leu enkephalin(0.1 nmol·L -1 ), IL-1α gene expression induced by LPS was inhibited. This inhibitory effect of enkephalins on LPS-induced IL 1α gene expression in the cultured hippocampal gliacytes could be blocked by an opioid receptor antagonist, naloxone. The results suggest that the effect of enkephalin on the IL 1α gene expression induced by LPS in the cultured hippocampal gliacytes of neonatal rat may be mediated by opioid receptor on hippocampal gliacytes or neurons.
