摘要
目的建立四甲基偶氮唑盐微量酶反应比色(MTT)研究N-乙酰半胱氨酸(NAC)对亚硫酸钠(Na2SO3)小鼠脾淋巴细胞毒性影响的方法。方法以不同剂量浓度的Na2SO3和NAC与Na2SO3联合作用下对小鼠脾细胞染毒后,采用MTT法测定各样本在570 nm波长下的光密度值(OD值)。结果随着Na2SO3浓度的增加,OD值逐渐下降。Na2SO3与NAC高剂量的联合作用组与单独作用的高剂量组相比有统计学意义(P<0.05)。结论一定剂量的Na2SO3对小鼠脾淋巴细胞的生长有抑制作用。NAC具有抗氧化损伤的作用,它在一定程度上抑制Na2SO3的毒性作用。
Objective To build a method to study the toxicity of NAC to Na2SO3 lymphocytes. Methods The mouse spleen lymphocytes were exposed to different doses of Na2SO3 with or without NAC(10 mmol/L) ,and the OD values of samples were detected at 570 nm wave length by MTT method, Results The OD values of different Na2SO3 groups were decreased significantly with the increasing concentrations of Na2SO3, The OD values of 10 mmol/L Na2SO3 with 10 mmol/L NAC group was significantly different from the 10 mmol/L Na2SO3 group, P 〈 0.05, Conclusion Certain concentration of Na2SO3 can inhibit the growth of mouse spleen lymphocytes. NAC has anti-oxygenation effect,and can inhibit the toxicity of Na2SO3 on lymphocytes to some degree.
