摘要
以BCR-ABL融合蛋白氨基酸序列为基础,采用EMBOSS软件及DNAstar软件结合Hopp and Woods亲水性参数、Janin可及性参数、极性参数、柔韧性参数及二级结构方案对慢性粒细胞白血病BCR-ABL融合蛋白(p210^(BCR-ABL))融合区的B细胞表位进行预测。结果表明:BCR-ABL蛋白融合区位于B细胞表位内,该表位含有α-螺旋和无规则卷曲结构。BCR-ABL融合蛋白融合区B细胞表位预测的研究对应用合成肽抗原制备融合区单克隆抗体具有重要的指导意义。
To predict B cell epitope for the fusion region of BCR-ABL fusion protein (p210^BCR-ABL) in chronic myeloid leukemia, based on BCR-ABL fusion protein sequence, the B cell epitope for BCR-ABL fusion region was predicted by methods of EMBOSS program, DNAstar software, hopp and woods hydrophility, Janin accessibility, polarity, flexibility and secondary structure. The Results showed that the fusion region of BCR-ABL fusion protein (p210^BCR-ABL) probably located in a B cell epitope which contained a-helix and random coil structure. Prediction of the B cell epitope for the fusion region of p210^BCR-ABL is helpful for preparation of monoclonal antibodies to BCR-ABL fusion region using synthetic peptide approach.
